This antibody recognizes protein of 192 kDa, which is identified as Clathrin Heavy Chain. Clathrin is composed of three heavy chains and three light chains, which associate non-covalently to form a triskelion structure. Clathrin heavy chain (HC) is composed of a terminal globular domain, a distal segment and a proximal segment containing a light chain-binding site. Reacts with a 75 kDa melanocyte-specific gene product, identified as Tyrosinase-related protein-1 (TRP-1). It is involved in melanin synthesis. TRP1 is present on the melanosomal membranes of melanoma, normal melanocytes and nevi. Recent evidence suggests that TRP-1 is involved in maintaining stability of tyrosinase protein and modulating its catalytic activity. TRP-1 is also involved in maintenance of melanosome ultrastructure and affects melanocyte proliferation and cell death. This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. Recognizes a protein of 130 kDa, identified as Desmoglein-3 (DSG3). This MAb is highly specific to Desmoglein-3 and does not cross-react with other members of the Desmoglein-family. DSG3 is a calcium-binding transmembrane glycoprotein component of desmosomes in vertebrate epithelial cells. Research has shown that DSG3 has a very high sensitivity (80%) and specificity (100%) in recognizing squamous cell carcinoma (SqCC). Therefore, DSG3 is considered a very important marker for lung SqCC and can be a useful ancillary marker to separate SqCC from other subtypes of lung cancer. Stem cell tyrosine kinase (STK-1) has been cloned from a CD34 hematopoietic stem cell enriched library and identified as the human homolog of a previously identified gene of Mouse origin designated either Flk-2 or Flt-3. The STK-1 cDNA encodes a protein of 993 amino acids with 85% identity to Flt-3/Flk-2. STK-1 is a member of the type III receptor tyrosine kinase family that includes Kit (steel factor receptor), Fms and PDGF. STK-1 expression in blood and marrow is restricted to CD34 cells, a population greatly enriched for hematopoietic stem/progenitor cells. STK-1 antiserum recognizes two polypeptides in these cells. The Mouse homolog of STK-1, designated Flt-3/Flk-2, is expressed at high levels in hematopoietic cells and also in neural, gonadal, hepatic and placental tissues. It is highly specific to VEGF, which is a homodimeric, disulfide-linked glycoprotein with a close homology to platelet-derived growth factor (PDGF). Thyroperoxidase (TPO) is a 933-amino acid, type I transmembrane glycoprotein that plays a key role in thyroid gland function and autoimmunity. It is present as a dimer on the apical surface of thyroid follicular cells.
